“文革”初期，杨贵首当其冲遭受冲击。他回忆道：“‘文化大革命’一起来，安阳地区的县委书记中，我是最早受到冲击的……（造反派的）街头大标语：‘大好消息，东扒焦裕禄的墓，西罢杨贵的官。’焦裕禄努力改变兰考面貌，我们自力更生修红旗渠，都成了‘罪恶’。”
据不完全统计，杨贵先后遭到批斗270多场。大批的林县干部群众、劳动模范和杨贵的家人都在所难免，受到牵连。红旗渠配套工程建设也被迫停滞。
1966年9月21日凌晨，安阳地委造反派星夜围攻林县县委揪斗杨贵。闻讯后，李先念马上做出批示，陶铸、谭震林即刻向周总理汇报。周总理让电话通知河南省委：请杨贵同志到北京参加国庆观礼以便保护他。遗憾的是，周总理的指示被地委造反派扣押，未能落实。
已任中南局第一书记的王任重也想保护杨贵，明确表态不同意杨贵回安阳地委“参加运动”。河南省委代理第一书记文敏生执行了王任重的指示。据他1967年在文字材料中所述：省委两次明确表示“杨贵可以不回地委”，而省委候补书记兼文革领导小组副组长戴苏理却擅自“答应可以。”这就极大地助长了造反闹事者的嚣张气焰。
万般危急之际，林县的广大干部群众不顾个人和家庭的安危，奋不顾身自发加入了保护杨贵、保卫红旗渠的行列。
1967年5月30日，已被关押、揪斗半年多的杨贵在陈亦庆、白兰香等北京大学生和林县工人们的帮助下，藏在装满棉花籽的大卡车内辗转到了山西。全国劳动模范李顺达和申纪兰义无反顾地收留保护了他。
6月10日，杨贵到了北京，先后在新华社记者方徨、程竟明、国务院外事办干部李后和京郊618军工厂工人师傅等好心人的家中躲藏，“像当年打游击和敌人周旋的战斗生活一样。”杨贵的入党介绍人王青奇、刘树杰夫妇也千方百计地竭尽所能帮助他。
